I have to say that it was Sellevision by Augusten Burroughs. All of his books are absolutely hilarious, especially Possible SIde Effects and Magical Thinking. He's just too funny for words, sometimes.

I don't read many funny books. However, the funniest I've ever read was Janet Evanovich, Stephanie Plume series. It actually made me laugh out loud.

Freedom and The Corrections - Jonathan Franzen
Wonder Boys - Michael Chabon
The World According to Garp and a Prayer for Owen Meany - John Irving
Any novel by Kurt Vonnegut.
These are subtly funny so it's hard to pick just one.

Good Omens by Neil Gaiman and Terry Pratchett. I've read it about 5 times and I still laugh every time I read it.

I've just read 'Futile Deceptions' and it's follow on 'Just Recompense' they are just out and I laughed most of the time. I don't know if it is in print but is on Kindle. Written by Douglas Wallis
